MANA 2026: Development of a Software-Defined Workflow using HILIC-HRMS with Targeted Quantitation of a Broad Panel of Polar Metabolites

To improve the efficiency of metabolomics labs and extend metabolomics into new labs, Move Analytical has been developing software-defined workflows that combine a physical kit — internal standards, external calibration and QC samples, extraction reagents, and a plate stack — with a software package designed to automate system suitability testing, experimental design, real-time data checks, automatic file processing, and result visualization. Having proven this approach with a CE-MS method (MoveKit™ CE) and a reversed-phase LC-MS method targeting bile acids (MoveKit™ BA), this work explores whether it extends to a broad polar metabolite panel using HILIC-MS.

The prototype MoveKit™ HILIC kit builds on the MoveKit CE standards, adding an early-eluting, strongly-ionizing internal standard for negative ESI, and pairs them with a targeted library of roughly 500 polar metabolites in MoveApp™. Using a 12-minute gradient on a BEH amide column with an Exploris 240, the workflow demonstrated reliable detection of internal standards and resolution of closely-eluting amino acid isomers in both positive and negative ionization modes, with initial testing underway in Move Analytical’s lab and external collaborator labs.
