Bile acids (BAs) have emerged as important biomarkers and signaling molecules implicated in diseases ranging from metabolic syndrome and inflammatory bowel disease to neurodegeneration, but reliable BA measurement remains technically challenging. Triple quadrupole pseudo-MRM assays suffer from high chemical background due to poor fragmentation, closely-eluting isomeric pairs are prone to software misassignment, and the rapidly expanding universe of conjugated BA species demands analytical flexibility beyond fixed target lists.
MoveKit™ BA (Move Analytical LLC) addresses these challenges by combining sample prep, experimental design, data collection, and data processing into a fully automated software-defined workflow. An Early Adopter Program assessed interlaboratory reproducibility across 6 laboratories on 7 Orbitrap and Stellar MS platforms, covering 32 stable isotope-labeled standards and 183 total BA targets. Across all experiments, MoveApp™ correctly assigned the 6 critical bile acid pairs in every case that passed the data check, with bile acid concentrations determined across 6 laboratories in 3 different matrices — outperforming iRT and explicit retention time approaches.
