MoveTip #2: Skyline: Enabling and Viewing Calibration Curves

⚡ Brief Summary In this MoveTip, we share how to set up calibration curves for quantitative MS analysis in Skyline software. A common question is: Why is my calibration curve not showing up in Skyline? This tip gives the two most common reasons that a calibration curve is not visible once your LC-MS data is loaded into Skyline, and how to fix it.

🧪 What You’ll Learn We heavily use Skyline Software for method development and in-depth data analysis tasks. Here we share with you a quick shortcut for: • Checking molecule settings to enable calibration curve calculations • Using the document grid to assign replicates appropriately for calibration

💻 Try It Yourself Here is how you can test this yourself right away: • Go to Settings / Molecule settings and set the Regression Fit in the Quantification Tab • Click View / Live Reports / Document Grid • Go to Reports / Replicates • Assign your calibration standards as “Standards”, and Assign the calibration values in the Analyte Concentration field. • If your calibration curve is not showing at all, go to View / Calibration Curve (not shown in video)
