MoveTip #6: Viewing raw mass spectral data directly in Skyline
⚡ Brief Summary Extracted ion chromatograms are used for quantification in LC-MS/MS, meaning a single ion or mass value extracted over the course of the chromatogram. However, due to the high dimensionality of mass spec data, it’s often useful to look at the full spectrum. In this MoveTip, we demonstrate using Skyline to view the spectrum without ever leaving the quantitative software package.
🧪 What You’ll Learn to try It Yourself You’ll learn how to use Skyline to: • View the full mass spectrum at any timepoint, for your HRMS instrument • Pan left and right across a peak in order to understand other molecules and interferences that may be coeluting • Zoom in and view the spectrum as profile or centroid, to see if the extracted ion chromatogram approach is matched to the raw data
